本试验以兔眼蓝莓主栽品种‘园蓝’为试材,研究其离体培养增殖技术。基本培养基为WPM培养基,添加蔗糖20g/L,琼脂10g/L,pH为5.2。实验总体分为两个过程:第一步,在基本培养基中加入ZT (2mg/L, 4mg/L, 6mg/L)或2ip (2mg/L, 5mg/L, 10mg/L),共6个处理,培养60d后,观察记录蓝莓腋芽萌发情况,WPM ZT 4mg/L更有利于腋芽萌发,各处理中腋芽萌发后生长速度都很慢;第二步,在基本培养基中加入4mg/L的ZT及IAA(0.1mg/L, 0.2mg/L)或IBA(0.1mg/L, 0.2mg/L),共4个处理,培养60d后进行数据统计,发现最佳处理为WPM ZT 4mg/L IBA 0.2 mg/L,并且芽子长势较好。因此,兔眼蓝莓‘园蓝’腋芽诱导和生长的最佳培养基为WPM ZT 4mg/L IBA 0.2 mg/L。 |

This work studied the in vitro multiplication of rabbit eye blueberry Garden Blue. The basic medium used is WPM plus sucrose 20g/L, agar 10g/L, with pH of 5.2. Two experiments were performed as follows. One was carried out as WPM plus ZT (2mg/L, 4mg/L, 6mg/L) or 2ip(2mg/L, 5mg/L, 10mg/L). Data were collected after 60 days. The results indicate that the best treatment for shoot formation from axillary buds is WPM plus ZT 4mg/L, shoots formed from all treatments grow very slowly. Another was performed as WPM 4mg/L of ZT supplemented with IAA (0.1mg/L, 0.2mg /L) or IBA (0.1mg/L, 0.2mg/L). 60 days later, we found that the best treatment for shoot formation is WPM ZT 4mg/L IBA 0.2 mg/L, and those shoots grow well. As a result, the best medium for shoot formation and growth from axillary buds of Garden Blue is WPM ZT 4mg/L IBA 0.2 mg/L. |
