【目的】为香蕉种质资源的鉴定、分类、保存和新品种选育提供科学依据。【方法】应用表型性状和SSR分子标记对20份香蕉材料进行了遗传多样性分析。【结果】表型聚类结果表明,供试材料可分为3组分别是香牙蕉组、粉蕉组和大蕉组,香牙蕉组因假茎的高度差异,进一步细分成3个亚组。13对多态性较好的SSR引物在这些品种中共产生525个条带,平均每对引物产生6.08个多态性条带,产物片段大小在99bp到360bp之间。利用Jaccard系数评估了品种间的相似性,这些香蕉品种的相似性数值在0.48到1之间。用UPGMA进行聚类分析,可将供试材料分为3组分别是香牙蕉组、粉蕉组和大蕉组,其中香牙蕉组的遗传多样性较低。本研究所用的13对SSR引物不能将所有材料区分开。【结论】香蕉种质资源丰富,同一组内部分材料间遗传相似性非常高,对其进行分类需将形态特征和分子标记两者有机结合起来。 |

[Objective]To provide scientific basis for the identification, classification, preservation and new varieties breeding of banana germplasm.[Method] Phenotypic characteristics and SSR molecular markers were used to analyze the genetic diversity of 20 banana germplasm materials. [Results]Phenotypic clustering showed that all the materials were divided into three group, Cavendish group, Fenjiao group and Dajiao group; the Cavendish group base on different pseudostem height, further subdivided into 3 subgroups. 13 SSR primers generated 525 polymorphic bands, with a mean number of 6.08 bands per pairs primer, the sizes of PCR products were between 99 bp to 360 bp. The genetic similarity between the accessions ranged between 0.48 and 1, which was estimated by Jaccard’s coefficient.According to the UPGMA method based on genetic similarities,materials were divided into Cavendish group, Fenjiao group and Dajiao group in which the Cavendish group had lower degrees of genetic differentiation relatively. All the tested cultivars could not be discriminated by the 13 primers. [Conclusion] Banana germplasm resources were rich, and the same group genetic similarity was very high. In order to determine the relationship among those species, we had better to combine the phenotypic characteristics and molecular markers for comprehensive analysis. |
