为了解chsV基因在尖孢镰刀菌古巴专化型侵染香蕉过程中的作用,及其与尖孢镰刀菌古巴专化型l号生理小种和4号生理小种之间的致病力差异的关系。采用PCR和RT-PCR方法扩增了2个生理小种的chsV基因,测序后对该基因的核苷酸序列和编码的蛋白进行分析。结果表明2个生理小种chsV基因不存在内含子,开放阅读框均为726bp,核苷酸同源性为99.6%,编码242个氨基酸,氨基酸序列相同。根据生物信息学软件预测编码蛋白没有信号肽,分子量为26346.8u,pI为6.18。2个生理小种寄主选择性差异与chsV基因并无明显对应关系,这为进一步研究chsV基因功能奠定了基础。 |

Fusarium oxysporum f.sp.Cubense is the pathogen of the Panama disease. In order to know the roles of chsV gene in invasion process of Foc into banana and its relationship in pathogenic difference between the races 1 and 4 of Foc attacking the same banana cultivar. the chsV genes were amplified by Polymerase Chain Reaction and Reverse Transcription Polymerase Chain Reaction, The amplified products were cloned and sequenced, and sequence alignments were done by using the biological softwares. the open reading frames of chsV from Foc1 and Foc4 were both 726 bp, encoding 242 amino acids with 26346.8u of Molecular weight and 6.18 of pI. The chsV gene from Foc1 showed 99.6% similarity to Foc4, but their amino acid sequences were the same. it seemed there was no relation between the different pathogenicity to the same banana variety and chsV gene on the face of the sequences of chsV genes and amino acid sequences. These results would help to know the function of the chsV gene from Foc1 and Foc4 in the future researches. |
