利用ISSR标记技术对14份香蕉种质的遗传多态性进行了分析。从15个ISSR引物中筛选出12个多态性引物,共扩增出126条DNA条带,其中多态性条带有101条,占80.2%。鉴定出4条特异出现DNA片段和7条特异缺失DNA片段。依据相似系数0.88的水平,将14份香蕉种质划分为4大类,与传统形态学分类结果相同。香牙蕉的遗传多样性较低,新选育的抗病突变型与母本之间不具有紧密的亲缘关系,不同香蕉种质对枯萎病的抗性强弱与其遗传相似性之间没有明确的相关性。本研究为香蕉品种的鉴定分类及亲缘关系的建立提供理论基础。 |

The genetic polymorphism of 14 banana germplasms was analysed using ISSR markers.Twelve primers were screened from fifteen arbitrary primers,and a total of 126 DNA bands were amplified,among which 101 (80.2%) were polymorphic.Four specific appearance DNA fragments and seven specific lackness DNA fragments were identified.14 banana germplasms were grouped into 4 clusters based on the similarity coefficient of 0.88,which was the same as the results of traditional morphological classification.The Cavendish bananas had lower degrees of genetic diversity,and there appeared no close genetic relationship between the novel disease resistance mutant types and female parent.The disease resistance of banana germplasms to Fusarium oxysporum f.sp.cubense race 4 had no clear relationship with their genetic diversities.This study provided the theoretical basis for the identification and classification of banana variety,and the banana genetic relationship building. |
