苏州地区宽皮柑桔的遗传多样性分析

      为研究苏州太湖地区及长江流域的桔类资源的遗传多样性,本研究利用EST-SSR、Indel分子标记对38份采自苏州太湖地区及长江流域的桔类资源进行了遗传多样性分析。从41对设计的EST-SSR和Indel引物中筛选出20对多态性强、重复性好的引物,对38份桔类种质DNA进行了扩增,对多态性位点进行了统计,采用结构分析、主成分分析和聚类分析方法,对不同材料的亲缘关系进行了分析。利用20对引物扩增,共获得84个多态性标记位点,240条多态性条带,平均每对引物检测到12个扩增条带。38份柑桔品种的观测等位基因数(Na)、有效等位基因数(Ne)、观测杂和度(Ho)、期望杂和度(He)以及香农信息指数(I)分别为3.63,2.06,0.215,0.435,0.79。结构分析、聚类分析和主成分分析揭示了苏州太湖地区以及长江流域古老宽皮柑桔的亲缘关系,结果表明黄皮桔和朱桔是两个古老的桔类资源,长江上游流域的红桔是与朱红桔不同的古老桔类资源。分析揭示了城固冰糖桔、大头料红、迟红为杂种起源类型,南桔、福桔与黄皮桔的亲缘关系较近,土桔、土柑为黄皮桔和朱红桔的杂种类型。研究结果为全面评价太湖地区桔类资源的遗传背景,鉴定桔类品种的亲缘关系和起源提供了重要参考,为今后桔类资源的保护利用及遗传育种研究提供了重要基础资料。

      To study the genetic diversity of mandarin landraces origin in basin of Tai Hu lake of Shu Zhou and Yangzi river area, we used expressed sequence tag-simple sequence repeat (EST-SSR) and insertion and deletion mutation (Indel) markers to reveal the genetic diversity of 38 citrus accessions from these area.20 pairs of SSR and Indel primers with polymorphic and stable bands were chosen from 41 tested pairs and used to amplify genomic DNA of 38 mandarin accessions, the polymorphic bands were counted and Structure analysis, principal component analysis as well as cluster analysis were applied to reveal the phylogenetic relationship of these citrus germplasms.A total of 240 polymorphic bands corresponding to 84 loci were recorded, with an average of 12 polymorphisms observed per primer pair. The mean value of observed number of alleles (Na), effective number of alleles (Ne), observed heterozygosity (Ho), expected heterozygosity (He) and Shannon’s index (I) were 3.63, 2.06, 0.215, 0.435, 0.79 respectively. Structure analysis, cluster analysis and principal component analysis were conducted to reveal the phylogenetic relationship of these mandarin accessions.It was concluded that Huang Pi Ju and Zhu Hong Ju are two groups of primitive mandarin accessions, Hong Ju which widely distributed in upper reach of Yangzi river is distinct from Zhu Hong Ju. The study also showed that the genetic background of Cheng Gu Bing Tang Ju, Da Tou Liao Hong, Chi Hong are complexity due to the introgression from different mandarin types. Nan Ju and Fu Ju have close relationship with Huang Pi Ju. Tu Ju and Tu Gan might arise from Huang Pi Ju and Zhu Hong Ju. This study unraveled the genetic background of some important mandarin accessions and identify their phylogenetic relationship, this will benefit to the preservation, utilization of mandarin germplasms as well as breeding program.
