      真龙柚是沙田柚的优变芽系选育而成。为了进一步确定芽变系性状的可遗传性,对沙田柚和真龙柚及其无性系子代6个样品进行了RAPD和SSR分析。结果显示:8个RAPD引物共扩增出43条稳定清晰条带,其中5条具有多态性,多态性比率为11.63%,平均每个引物扩增5.37条带;26对SSR引物共扩增出34条带,只有5条带具有多态性,多态性比率为14.71%。结果表明:真龙柚与沙田柚的遗传距离为0.063,它们之间在DNA水平上存在遗传差异,说明真龙柚是由遗传物质改变引起的,非环境因素控制,具有可遗传性,通过RAPD和SSR技术可以有效鉴定,RAPD引物A07、A05和SSR引物SS15、TAA27均能将真龙柚和沙田柚鉴别出来。

      Zhenlong pummelo is a high-quality bud mutation line selected from Shatian pummelo cultivar. The six samples of Shatian pummelo and generation clone of Zhenlong pummelo were analyzed using RAPD and SSR to affirm the character hereditability of bud mutation further. Zhenlong pummelo is a high-quality bud mutation line selected from Shatian pummelo cultivar. The six samples of Shatian pummelo and generation clone of Zhenlong pummelo were analyzed using RAPD and SSR to affirm the character hereditability of bud mutation further. The results showed that 43 bands were amplified from 8 random primers, among which 5 bands were polymorphic bands, and the polymorphic percentage was 11.63%, 5.37 bands were obtained per primer in average; 34 bands were amplified from26 SSR primer pairs, among which 5 bands were polymorphic bands, and the polymorphic percentage was 14.71%. The genetic distance between Zhenlong pummelo and Shatian pummelo was 0.063,and there was difference in DNA between them, The results indicated that there were some molecular level differentiation between them; and the differentiation could be identificated by RAPD and SSR, Zhenlong pummelo and Shatian pummelo could be identificated by RAPD primer (A07 and A05) and SSR primer (SS15 and TAA27).
