摘要:为了给甜樱桃微生物肥料的研制和生产提供优良菌株资源。本文从马哈利樱桃根际土壤中分离获得52株细菌,通过保绿法和萝卜子叶增重法进行IAA产生菌的筛选,并经形态观察、生理生化特征及16S rDNA序列分析明确分类地位,利用分光光度法测定产IAA的能力,并对最佳碳氮源、产素条件进行了筛选。结果表明52株菌中B20产IAA能力最强,经鉴定为阿氏芽孢杆菌(Bacillus aryabhattai),其最佳产素的培养条件为:采用10%的装液量在温度为30℃、转速为150 r? min-1的条件下培养40h,最佳碳、氮源分别为果糖和酵母粉。首次研究发现阿氏芽孢杆菌具有IAA产生能力,菌株B20具有作为功能微生物肥料研发的潜力,可进一步对其进行开发研究。 |

Abstract:The paper aimed at providing the excellent strains for the development and production of cheery microbial fertilizer. 52 bacteria strains were isolated and purified from the rhizosphere soil of Mahaleb. IAA biosynthesis strains were screened using the methods of remaining green and radish cotyledon weight dioassay. And the strains were identified through the morphological feature and 16S rDNA sequence analysis. Among them, strain B20 was the highest in IAA excreting capability. It was identified as Bacillus aryabhattai. The conditions for the strain growth and fermentation were optimized by measuring IAA content. The optimal culturing conditions for both growth and IAA secretion were filling volume 10%, incubation at 37℃ for 44h and shaking speed 150 r? min-1, fructose and Yeast powder as carbon source and nitrogen source respectively. The strain B20 is expected to be excellent microbial fertilizer. |
