利用实时荧光定量PCR对香蕉枯萎病抗感品种受枯萎病菌浸染后不同时间内的10个防卫或潜在防御相关基因进行表达分析。结果表明,接种Foc4后,10个基因在抗病品种‘宝岛蕉’和感病品种‘巴西蕉’中均有表达,但其表达模式有差异。接菌后1~192 h,漆酶(laccase)4B、谷胱甘肽硫转移酶(glutathione-S-transferase Cla47)(9 h和120 h除外)、Class III型过氧化物酶(class III peroxidase 136 precursor)、类几丁质酶(putative chitinase)和纤维素合成酶催化亚基(cellulose synthase catalytic subunit 12)等5个基因在抗病品种‘宝岛蕉’中的表达水平均高于感病品种‘巴西蕉’。此外,接菌后除个别时间点外,漆酶4A、周质空间β-葡萄糖苷酶前体合成酶(periplasmic beta-glucosidase precursor)、过氧化物酶(peroxidase N)、类似烟草叶片表面抗性蛋白(similar to T-phylloplanin)和抗菌肽(snakin-1)等5个基因在‘宝岛蕉’中的相对表达量却均低于‘巴西蕉’。 |

Real-time quantitative PCR was used to determine expression levels of ten banana defence-related genes or potential defence-responsive genes after resistant cultivar ‘Formosana’ and susceptible cultivar ‘Brazilian’ roots infected by Fusarium oxysporum f. sp. cubense in different times. The results showed that expression of the ten genes was detected in ‘Formosana’ and ‘Brazilian’, but the expression patterns of these genes was differential. The expression levels of glutathione-S-transferase Cla47 (9 h and 120 h excepted), laccase 4B, class III peroxidase 136 precursor, putative chitinase and cellulose synthase catalytic subunit 12 were higher in the resistant cultivar ‘Formosana’ than in the susceptible cultivar ‘Brazilian’ after the seedling infection by Foc4. In addition, the expression levels of laccase 4A, periplasmic beta-glucosidase precursor, peroxidase N, similar to T-phylloplanin and snakin-1 were lower in the resistant cultivar ‘Formosana’ than in the susceptible cultivar ‘Brazilian’ after the seedling infection by Foc4. |
