      柑橘黄龙病病菌是专性寄生在柑橘韧皮部筛管组织中。为了探索柑橘韧皮部蛋白(Phloem Protein 2, PP2)在寄主与黄龙病互作中的功能,本实验以高感品种锦橙和耐病品种酸柚病枝上的叶片为材料,通过实时荧光定量PCR的方法从全基因组水平初步筛选到6个响应黄龙病侵染诱导的韧皮部蛋白基因PP2。并比较了其在高感品种锦橙和耐病品种酸柚中的相对表达量。结果发现CsPP2B15可能与耐病有关,CsPP2B2、CsPP2B10可能与感病相关。采用生物信息学的方法对其编码氨基酸序列从结构组成、基本理化性质、亲、疏水性进行分析,构建了系统发育树比较发现其与大豆和拟南芥中PP2基因家族的亲缘关系较近。为下一步挖掘潜在的黄龙病抗、感基因进行抗性分子育种提供了理论依据。

      Candidatus Liberibacter asiaticus “Las” associated with citrus Huanglongbing disease is a phloem-limited bacterial pathogen. To explore functions of Phloem Protein 2(PP2) in the interaction between Las and host plants, expression profiles of PP2 genes were investigated using real-time fluorescence quantitative PCR (qPCR) in this study. Relative expression analysis of 109 PP2 genes in susceptible variety Jincheng showed that six PP2 genes (CsPP2B15, CsPP2B12-1, CsPP2B10, CsPP2B12-2 and CsPP2B2, CsPP2A) significantly responded to HLB infection. Expression levels of the six genes were further evaluated between susceptible variety Jincheng and resistant variety sour pomelo. The results showed that PP2B15 was related to citrus disease resistance while PP2B2 and PP2B10 may be associated with host susceptibility. The bio-informatics method was used to analyze the amino acid sequence from the structure, basic physical and chemical properties, affinity and hydrophobicity. Phylogenetic analysis showed that the six CsPP2 genes had a close relationship with PP2 family genes from soybean (Glycine max) and Arabidopsis. The study offers some theoretical basis for citrus HLB resistance molecular breeding.
