      以山定子和Y系早花山定子为试材,分析苹果不同花发育相关基因MbFT1、MbFD、MbMADS12、MbAP1、MbSOC1在山定子和早花山定子的芽及韧皮部中的表达模式,以期为探究早花山定子早花机制提供理论依据。研究结果显示,MbAP1基因在山定子和早花山定子中的表达模式及表达量可能是影响早花山定子早花发育的一个重要因素,分别以山定子和早花山定子的基因组DNA为模板,扩增MbAP1基因的启动子序列。研究结果显示,山定子和早花山定子MbAP1基因的启动子序列存在碱基差异位点,其中在-388bp碱基处碱基由C 变为G,作用元件由山定子的生长素响应元件(TGA-element)变为早花山定子的水杨酸响应元件(TCA-element),推测MbAP1基因在山定子和早花山定子中表达模式的不同可能与两者启动子碱基差异有关。
英文摘要:
      The expression patterns of several floral development genes, including MbFT1, MbFD, MbMADS12, MbAP1 and MbSOC1, were investigated in the bud and phloem of Malus baccata and early blooming Y series Malus baccata. The results showed that the expression patterns and expression levels of MbAP1 gene in Malus baccata and early blooming Malus baccata may be an important factor affecting early flower development. The genomic DNA of Malus baccata and early blooming Malus baccata were used as the templates to clone the promoter of MbAP1, respectively. The results show that there are base differences in the promoter sequence of MbAP1 gene between Malus baccata and early blooming Malus baccata. When the base at -388 bp changed from C to G the acting element changed from the auxin- responsive element (TGA-element) of the Malus baccata to the salicylic acid responsive element (TCA-element), it is speculated that the difference of the expression pattern of MbAP1 gene between Malus baccata and early blooming Malus baccata may be related to the differences of the bases of the two promoters.
