从香蕉果肉中克隆得到了一个类黄酮-3-O-葡糖基转移酶基因UFGT1基因全长,利用生物信息学方法分析了该基因的基本理化性质、蛋白二级结构、亚细胞定位、亲疏水性、信号肽、跨膜区、磷酸化位点、基因结构、保守结构域、系统进化关系,并对该基因的时空表达模式及香蕉果实不同发育阶段进行了表达分析。结果表明:UFGT1基因长1380bp,编码459个氨基酸的蛋白质;该蛋白为疏水性蛋白,亚细胞定位在叶绿体,蛋白无跨膜结构,不存在信号肽;MaUFGT1在不同组织中均有表达,但表达丰度存在差异;MaUFGT1在‘巴西’和‘香粉1号’两个不同品种中,随着果实成熟,其表达量逐渐上升,后期‘香粉1号’明显高于‘巴西’。这些结果表明,MaUFGT1可能参与了香蕉果肉黄酮类物质的合成,为进一步研究MaUFGT基因的功能奠定了基础。 |

In this study, a flavonoid-3-O-glucosyltransferase gene(UFGT), was cloned from bananas. The basic physicochemical properties, protein secondary structure, subcellular localization, hydrophobicity, signal peptide, transmembrane region, phosphorylation site, gene structure, conserved domain and phylogenetic relationship of the gene were analyzed by bioinformatics method. The expression pattern of the gene and the different developmental stages of the banana fruit were analyzed. The results showed that the UFGT gene was 1380 bp, which encoded 459 amino acids. The protein was hydrophobic and the subcellular was located in the chloroplast. Transmembrane structure and no signal peptide was not present in the protein. MaUFGT1 was expressed in different tissues, and the expression abundance was different. Expression of MaUFGT1 in the ‘BaXi’ and ‘ xiangfen 1’banana gradually increased during the fruit develomental period, and expression of MaUFGT1 in the‘ xiangfen 1’was significantly higher than ‘BaXi’ in the later period. The results showed that MaUFGT1 might be involved in the synthesis of flavonoids in fleshy pulp, which lays a foundation for further study of the function of MaUFGT gene. |
