由柑橘黄单胞菌杧果致病变种(Xanthomonas citri pv.mangiferaeindicae, Xcm)引起的杧果细菌性角斑病是杧果上的一种重要细菌性病害,严重危害杧果产业发展。为了研究抗毒素phd基因的结构和功能,本试验通过在NCBI上选取已被注释的Xcm抗毒素phd基因的序列,设计出引物phd-F和phd-R,从Xcm003菌株中扩增出258 bp的DNA片段,提交至NCBI的登录号是MH401645。经序列分析该片段是一个完整的phd基因,编码85个氨基酸,其理论分子量9.68 kD,理论等电点为9.39,是一种亲水性蛋白,无信号肽,有6个磷酸化位点以及一个结构域。系统进化树结果显示,该基因的氨基酸序列与Xanthomonas citri pv.mangiferaeindicae LMG941的Phd蛋白聚为一类。通过克隆杧果细菌性角斑病菌phd基因并对其进行生物信息学分析,为深入研究phd基因做基础。 |

The bacterial leaf spot pathogen of mango which is caused by Xanthomonas citri pv. mangiferaeindicae is one of the important bacterical diseases, endangering the mango industry seriously. To research the structure and fuction of antitoxin phd gene. In this study, primers phd-F/phd-Rwere designed according to the annotated Xcm antitoxin phd gene sequence on NCBI, and from sequence analysis, the 258 bp DNA fragment, augmented from Xcm003, was a complete phd gene (MH401645), encoding 85 amino acids and it’s molecular weight and theoretical isoelectric point are 9.68 kD and 9.39 respectively, it’s a hydrophilic protein with no signal peptide, this sequence contains six phosphorylation sites and one conserved domain. Phylogenetic analysis indicated that the amino acid sequence of this gene showed 100% high similarity with Phd protein of the Xanthomonas citri pv.mangiferaeindicae LMG941. By cloning the phd gene of the pathogenic bacteria of bacterial leaf spot pathogen of mango and carrying out bioinformatics analysis to it, we can make a preparation for a further research of phd gene. |
