土壤是制约苹果根系生长的主要因素。为了进一步分析苹果砧木根系对碱胁迫的响应,首先对两种耐碱性不同的苹果砧木幼苗——富平楸子(Malus prunifolia)(耐碱性强)与平邑甜茶(M. hupehensis)(耐碱性较弱)的根系结构和脱落酸(ABA)含量进行了分析。之后,又对富平楸子进行了转录组分析。在碱胁迫15 d后,和富平楸子相比,平邑甜茶幼苗的根系生长明显降低。两种砧木幼苗根系中脱落酸(ABA)含量均在48 h增加,但平邑甜茶根系中ABA含量的增加幅度较小。qRT-PCR分析结果显示,在富平楸子中,CIPK1和AHK1表达量在24 h和48 h上调。而在平邑甜茶中,AHK1表达量在48 h有缓慢的上升,CIPK1的表达量下调。这些结果表明,碱胁迫下,ABA含量的增加可能促进根系的生长,提高植物耐碱性。 |

Soil is the main factor restricting the growth of apple roots. In order to analyze the response of apple rootstock roots to alkali stress, Firstly, the root structure and abscisic acid (ABA) content of two apple rootstocks, Malus. prunifolia (alkali tolerant) and Malus. hupehensis (alkali sensitive), were analyzed. After that, M. prunifolia was analyzed by transcriptome analysis. After 15 days of alkali stress, the root growth of M. hupehensis was significantly lower than that of M. prunifolia. The content of abscisic acid (ABA) in roots of the both rootstocks increased at 48 h, but ABA content in roots of M. hupehensis increased slightly. The results of qRT-PCR analysis showed that the expression of CIPK1 and AHK1 was up-regulated at 24 h and 48 h in M. prunifolia. However, in M. hupehensis, the expression of AHK1 increased slowly and the expression of CIPK1 decreased at 48 h. These results suggest that the increase of ABA content under alkali stress may promote root growth and improve plant alkali tolerance. |
