研究了罗汉果花粉在超低温保存条件下的花粉寿命,并对罗汉果花粉的超低温保存技术程序进行了研究。结果表明:罗汉果超低温保存的程序为新鲜花粉脱水(低于含水量30%)后密封保存在超低温冰箱或液氮(LN)中,结束保存时采用冷藏、室温逐级化冻花粉,授粉前使用MTT染色法快速鉴定花粉生活力。 |

This paper focuses on the effects of cryopreservation on the pollen of siraitia grosvenorii and provides a promising cryopreservation procedures. The results showed that the procedure of cryopreservation of siraitia grosvenorii was the fresh pollen was dehydrated by anhydrous calcium chloride for 72 h (below 30% water content) and then sealed and stored in ultra-low temperature refrigerator (-86 ℃ ). At the end of storage, the frozen pollen was thaw out step by water bath(38℃) before use, and MTT staining can be used to test pollen viability quickly before pollination. It was simple, low cost and high pollen survival rate dealing with the method above. It will be used for large-scale planting of Siraitia grosvenorii with spray pollination technology. |
