| 长江上中游地区是世界李的原产地,李品种资源丰富,传统主栽品种为江安大白李；为了快速甄别重庆地方李与江安大白李之间的亲缘关系,加快重庆地方良种李新品种培育,该文采用DNA条形码技术,运用分子鉴定方法,从分子水平对重庆开州区选择的开州晚熟李与江安大白李进行matK、rbcL、ITS序列测定和比对,构建物种NJ进化树,快速鉴定两者之间的亲缘关系,指导地方李新品种培育。通过对开州晚熟李和江安大白李叶片样品的matK、rbcL、ITS序列测定,发现两种地方李的3种序列中,ITS、rbcL序列NJ进化树均表现为梳子状,差异不明显,两者之间达不到区分“种”的程度；matK序列NJ进化树之间有明显的差异。研究表明,开州晚熟李与江安大白李亲缘关系较近,属同种植物,但两者存在品种间遗传差异,通过进一步的培育有望育成新品种。 |

| The upper middle reaches of the Yangtze River is the origin area of Plum, and is of great Plum variety resources, among all the varieties in the area the Jiang"an big white Plum is the most planted traditional variety; To rapidly distinguish the phylogenetic relationship between Chongqing Local Plum and Jiang"an big white Plum, and to accelerate the seed-breeding of improved Chongqing local Plum varieties, this paper used DNA barcoding technique, as well as molecular identification methods, determied and compared the matK, rbcL, ITS sequence from molecular level between the Kaixian late-maturing plum and Jiang"an big white plum. We also constructed the species NJ phylogenetic tree, which can be used to rapidly identify the phylogenetic relationship between the former two species, and guide the cultivation of local Plum new species.Through the matK ,rbcL and ITS sequence determination on the leaves samples of Kaixian late-maturing plum and Jiang"an big white plum,we found that the Kaixian local plum and Jiang"an big white"s three sequences,the NJ phylogenetic tree of the ITS,rbcL sequences appears to be comb-like,the difference is not obvious enough to be distinguished as two different species;On the other hand,there are obvious differences between the two NJ phylogenetic tree for matK sequence the two trees. The studies have show the phylogenetic relationship between Kaixian late-maturing plum and Jiang"an big white plum is close and they belong to the same species,but they have the genetic differences between varieties,through further development it is highly possible to breed a new varieties. |
