      炭疽病是柑橘上发生普遍的一种病害,目前该病主要依赖化学防治,对柑橘产业的绿色发展造成了严重影响。本研究通过皿内对峙法和离体叶片接种法筛选到一株柑橘炭疽病生防内生细菌GN222,其对炭疽病菌的生长有较强的抑制作用,对炭疽病的发生有明显的延迟作用。通过形态特征、培养性状、生理生化特性测定和16S rDNA基因序列分析,将GN222归为芽孢杆菌(Bacillus sp.)。采用单因素法优化菌株GN222的摇瓶发酵条件发现,当GN222种子液按照5%的量接种到装有50mL、pH8的2×YT培养液的300ml三角瓶中,在30℃下以200 rpm振荡培养24h后,其发酵滤液对柑橘炭疽病菌的抑菌活性最强。试验结果可为菌株GN222的开发利用提供理论支持,并为柑橘炭疽病的生物防治奠定基础。
英文摘要:
      Anthracnose is a common disease in citrus. At present, the disease mainly relies on chemical control and has a serious impact on the green development of citrus industry. In this experiment, an endophytic bacterium named GN222 with the characteristics of inhibiting the growth of Colletotrichum gloeosporioides and delaying the occurrence of citrus Anthracnose was screened by in plate and on leaf in vitro. By testing of morphological, cultural,physiological, biochemical characteristics and the analyses of 16S r DNA sequence, GN222 was indentified as Bacillus sp.. The single factor method was used to optimize the fermentation conditions of strain GN222 in shaking flask. It was found that the fermentation broth had the strongest inhibitory effect on Colletotrichum gloeosporioides when the seed solution of strain GN222 was inoculated into 50mL culture medium with initial pH value of 8 in 5% quantity and the liquid was cultured in 2YT medium with the temperature of 30℃ and 200rpm shaking frequency for 24h. These results provided the theoretical supports for the application of strain GN222 and laid the foundation for biological control of citrus anthracnose.
