以‘徐香’猕猴桃种子为外植体,开展附加不同植物生长调节剂的培养基对种子萌发、增殖及生根培养的对比实验,筛选适合试管苗各阶段生长的最佳培养基配方,以期建立其实生苗快繁体系。结果表明,75%酒精消毒30 s,0.1% HgCl2消毒8 min为外植体最佳消毒方法,成活率达79.18%;种子最佳萌发诱导培养基为MS+100 mg.L-1肌醇;适宜的增殖培养基为MS+1.0 mg.L-1 6-BA+0.1 mg.L-1 NAA;生根培养基以1/2MS+0.4 mg.L-1 IBA最好;选用草炭土和珍珠岩(体积比4:1)移栽,植株根系包被苔藓,成活率达80%以上。 |

The seeds of ''Xuxiang'' kiwifruit were used as explants, Medium compounding proportion of different plant growth regulators were studied on seed germination, proliferation and rooting culture. The most suitable medium for rapid propagation in vitro was selected, and the rapid propagation system was established. The results showed that: the best disinfection method was treated with 75% alcohol for 30 s, then by 0.1% HgCl2 for 8 min, the survival rate was up to 79.18%; the best germination medium was MS+100 mg.L-1 inositol; at the multiplication stage, MS+1 mg.L-1 6-BA+0.1 mg.L-1 NAA was the best medium; 1/2MS+0.4 mg.L-1 IBA is the best rooting medium; transplanted with peat soil and perlite (4:1 by volume), plant roots coated with moss, the transplant survival rate reached more than 80%. |
