以广西富川大田栽培的脐橙根系为试验材料,探讨五种染色剂即苏丹红Ⅳ、酸性品红、苯胺蓝、台酚蓝和Quink牌纯黑墨水对脐橙根系丛枝菌根真菌染色效果的影响。研究结果表明:Quink牌纯黑墨水染色效果最佳,根系丛枝菌根真菌的菌丝、泡囊、丛枝等结构清晰可见。Quink牌纯黑墨水染色效果极佳、稳定不易褪色、操作简单、安全无毒,适用于脐橙根系丛枝菌根真菌的染色和制片观察。对广西富川脐橙不同采样点的根系进行染色,结果表明丛枝菌根在脐橙根系上普遍存在。 |

An experiment was conducted to evaluate the navel orange roots dyed with five stains i.e. Sudan red Ⅳ, acid fuchsin, aniline blue, trypan blue and Quink pure black ink. The results show that Quink pure black ink staining has the best effect, and the hyphae, vesicle, arbuscular and other structures of the AM fungal in the roots were clearly visible. The Quink pure black ink staining effect is the best, stable and not easy to fade, simple to operate, safe and and non-poisonous. Therefore is suitable for the staining and preparation of arbuscular mycorrhizal fungi in navel orange roots. Roots of navel orange in Fuchuan, Guangxi were dyed with Quink black ink. The results showed that arbuscular mycorrhiza was common in navel oranges. |
