为了解浙江省不同主产地脐橙的品质现状及亲缘关系,本研究对从5个栽培地引种的17个脐橙样品的理化品质性状进行分析,同时进行基因组DNA多态性分析, 从300个引物中筛选出具有丰富多态性的17个10bp的随机引物,共扩增出109个DNA片段,片段大小在200bp-2800bp之间,其中多态性谱带为65条,占59.63%,表现出较为丰富的RAPD多态性,根据遗传距离,利用UPGMA构建了品种亲缘关系树状图。结果表明,浙江省不同产地的脐橙品种固形物含量在12%-15.25%,朋娜维生素C含量较纽荷尔低,朋娜平均裂果率达14.48%,纽荷尔平均裂果率为4.04%,其它脐橙品系的裂果率平均为10.08%,浙江省纽荷尔脐橙总体品种特性较朋娜和其它脐橙品系佳。结合脐橙品质和遗传距离,将浙江省主产17份脐橙材料可聚成三大类群,Q001,Q002,Q003,Q006,Q007,Q0013之间的亲缘关系较近,为朋娜品种,Q0011,Q0015,Q0016,Q0017,可分为一类,为纽荷尔品种,其它品系的脐橙归为一类。 |

In this study, the quality characteristics of 17 navel orange samples introduced from 5 cultivation areas were analyzed to study quality status and kinship of navel oranges in different main producing areas of Zhejiang Province, at the same time, genomic DNA polymorphism analysis was performed, 17 10bp random primers which showed rich polymorphism were screened out from 300 primers. 109 DNA fragments ranging from 200 to2800 bp were amplified from this 17 random primers, of which 65 polymorphic bands, accounting for 59.63%, showed rich RAPD polymorphisms. According to genetic distance, A tree map of cultivar phylogeny was constructed by UPGMA. The results showed that the solid content of navel orange cultivars in different regions of Zhejiang Province ranged from 12% to 15.25%. The vitamin C content of penna was lower than that of Newhall. The average rate of fruit split was 14.48% , the average rate of Newhall was 4.04% and the the average rate of others was 10.08% . The quality of Newhall varieties is generally better than that of penna and other varieties. According to the quality and genetic distance of navel oranges, 17 navel oranges produced in Zhejiang Province can be grouped into three groups.Q001, Q002, Q003, Q006, Q007, and Q0013 from different origins have closer genetic distances and relationship, which are Penna. Q0011, Q0015, Q0016, and Q0017 which have closer genetic relationship are Newhall. |
