      本文以原产我国大别山区的两份完全雄性柿种质,‘雄株3号’、‘雄株8号’,以及偶开雄花的完全甜柿‘鄂柿1号’为试材,探讨不同浓度的蔗糖(100 g/L、150 g/L、200 g/L)、硼酸(100 mg/L、150 mg/L、200 mg/L)和硝酸钙(100 mg/L、150 mg/L、200 mg/L)对悬滴培养条件下花粉萌发的影响,同时比较碘-碘化钾(I2-KI)和TTC(2,3,5-氯化三苯基四氮唑)染色法在花粉活力检测效果上的差异。结果表明,蔗糖对柿花粉离体萌发的影响最大;这三种花粉在蔗糖含量为200 g/L、硼酸含量为150 mg/L、硝酸钙含量为100 mg/L时萌发率最高;I2-KI和TTC染色均可快速测定柿花粉活力,但前者测定需时更短;I2-KI和TTC染色检测结果均低于悬滴培养法。因此,花粉活力检测以不同技术相结合的方式为宜。本研究结果可为柿遗传改良中花粉活力的检测提供科学依据。

      In this paper, two complete male persimmon germplasms originating in the Dabie Mountains of China, ‘Male 3’ and ‘Male 8’, as well as PCNA ‘Eshi 1’, which even opens male flowers, were used as research materials to explore the influences with different concentrations of sucrose (100 g/L, 150 g/L, 200 g/L), H3BO3 (100 mg/L, 150 mg/L, 200 mg/L), and Ca(NO3)2 (100 mg/L, 150 mg/L, 200 mg/L) on pollen germination in vitro under hanging drop culture conditions, and compare the effect of I2-KI and TTC (2,3,5-triphenyltetrazolium chloride) staining methods on pollen vitality detection. The results showed that sucrose had the greatest effect on the persimmon pollen germination in vitro; Pollens germination rate of ‘Male 3’, ‘Male 8’ and ‘Eshi 1’ were highest when the sucrose contentration was 200 g/L, H3BO3 contentration was 150 mg/L, and the Ca(NO3)2 contentration was 100 mg/L. Both I2-KI staining and TTC staining could rapidly determine the pollen viability of persimmon, but the former takes the shortest time to determine. The two methods were significantly lower than that measured by the hanging drop method. Therefore, the combination of different techniques for pollen vitality detection is appropriate. The results of this study can provide a scientific basis for the detection of pollen vitality in the genetic improvement of persimmon.
