自田间采集沃柑黄化植株,盆栽在防虫温室中,二个多月后长出的第一批新梢即恢复绿色,但盆栽两年后再次出现黄化植株。选择盆栽的黄化植株和已恢复绿色的健康植株为材料,通过检测柑桔黄龙病菌、盆栽土壤的pH值和营养物质含量及根系的真菌分子种群,分析沃柑叶片黄化的相关因子,为沃柑黄化现象的综合治理提供依据。经PCR检测,盆栽沃柑健康植株和黄化植株均未感染黄龙病菌;健康植株根系发达,黄化植株根系相对较少。取除根尖外的褐色根段,针对真菌的ITS区域进行高通量测序,发现褐色根段中重要真菌种类是腐皮镰孢菌(Fusarium solani),其相对丰度在健康植株(60.347%)和黄化植株(47.069%)的根段间无显著性差异。盆栽土壤中的pH值及有机质、全氮、碱解氮、速效磷和速效钾的含量也无显著性差异。盆栽的黄化植株经重修剪枝条及断根处理后,长出的新梢叶片恢复健康绿色。可见,在没有感染黄龙病和土壤条件基本一致的条件下,沃柑植株的黄化现象主要是根系稀少及老化,影响了根系的吸收功能而引起的,断根处理可促进新根生长,提高吸收功能,进而使黄化植株新梢恢复健康绿色。 |

To provide a basis for integrated management Orah leaf yellowing, the factors related to Orah leaf yellowing were analyzed. Leaf yellowing and leaf healthy plants were choosed as materials for detecting Candidatus Liberibacter asiaticus (Las), pH and nutrient content in potting soils and fungal molecular population in brown root fragments. The results shown that Las was not detected in leaf yellowing and healthy plants of Orah. There were no significant difference in pH, content of organic matter, total nitrogen, alkaline hydrolysis nitrogen, available phosphorus and available potassium in potted soils between yellowing and healthy plants. Fusarium solani was the main fungus species with relative abundance of 60.347% and 47.069% in Orah brown root fragments of yellowing and healthy plants respectively, which was also no significant difference. The roots of healthy plants were well developed, while roots of leaf yellowing plants were less relatively. After heavy pruning and root-cutting, the new shoots of yellowing plants recovered to healthy green. Thus it can be seen, if Orah plants were not infected by Las and soil conditions were consistent basically, the cause of the yellowing phenomenon was mainly sparse and ageing of roots, which affect the absorption function of roots. The new shoots of yellowing plants could turn healthy green after root-cutting. |
