为了明确湖南省涟源市白马镇枣树叶斑病的病原菌种类,从涟源市白马镇采集枣树病叶10份。用组织分离法对采集的枣树病叶进行分离、纯化,采用形态学特征、rDNA-ITS、EF-1α基因进行菌株鉴定,利用分生孢子悬浮液喷洒法进行致病力测定。从枣树叶斑病中分离得到12株分离物,经过形态学特征、rDNA-ITS、EF-1α序列分析和致病力测定,证实枣树叶斑病的病原菌是葡萄座腔菌Botryosphaeria dothidea。实验验证了涟源市白马镇枣树叶斑病的病原菌是葡萄座腔菌B. dothidea。 |

In order to clarify the pathogen species of jujube leaf spot disease in Baima Town,Loudi City, Hunan Province, 10 samples of diseased leaves of jujube were collected from BaimaTown, Lianyuan City. The infected leaves of jujube were isolated and purified by tissue separation method. The strains were identified by morphological characteristics, rDNA-ITS and EF-1α genes. The pathogenicity of the bacteria was determined by conidia suspension spraying method. twelve isolates were isolated from leaf spot of Jujube. After morphological characteristics, rDNA-ITS, EF-1α sequence analysis and pathogenicity determination, it was confirmed that the pathogen of jujube leaf spot was Botryosphaeria dothidea. Experiment was confirmed that B. dothidea was the pathogenic bacterium of jujube leaf spot in Baima Town, Lianyuan City. |
