      本研究通过基因克隆和测序方法,对来自国内外Foc 的1号和4号小种的6个菌株endo-PG基因同源性、与其他真菌的亲缘关系以及endo-PG基因和氨基酸序列进行了分析,为香蕉枯萎病菌致病机制的研究奠定基础。结果表明,同一小种的endo-PG序列同源性高、亲缘关系近,不同小种的同源性较低、亲缘关系较远;总体来说,4号小种比1号小种同源性更高、亲缘关系更近。在全基因序列分析中,并没有发现明显的基因差异可以作为区分2个小种致病性差异的依据。根据预测的CDS序列翻译成氨基酸序列比对,发现6个菌株的endo-PG基因编码的氨基酸变异并没有引起相关保守结构域的变化,推测6个菌株的致病性可能与endo-PG基因的调控相关。
英文摘要:
      In this study, by means of gene cloning and sequencing, the homology of endo-PG genes of 6 strains from races 1 and 4 of Foc at home and abroad and their genetic relationship with the endo-PG genes of other fungi were analyzed, and their endo-PG genes and amino acid sequences were analyzed too, which laid a foundation for the study of the pathogenic mechanism of Foc. The results showed that the endo-PG sequences of the same race had high homology and close genetic relationship, while different races had lower homology and farther relationship. In general, sequence homology and the genetic relationship of race 4 strains were higher and closer than those of race 1 strains. In the whole gene sequence analysis, no obvious gene differences were found that can be used as the basis for distinguishing the pathogenicity differences between the two races. According to the translation of the predicted CDS sequence into amino acid sequence alignment, it was found that the amino acid variation encoded by the endo-PG gene of the 6 strains did not cause changes in the relevant conserved domains. It is speculated that the pathogenicity of the 6 strains might be related to the regulation of endo-PG gene.
