2021年,在海南保亭植物园发病的红毛丹(Nephelium lappaceum L.)活树上发现了一种灵芝根腐病。为明确该病害的病原菌种类及其生物学特性,笔者采用常规组织分离法,从其担子果分离出10株形态相同的菌株,并选取其中具有代表性的优势菌株 (编号为HMDGF001) 对其进行致病性测定、形态学特征鉴定、rDNA-ITS、SSU和LSU序列分析,构建系统发育树以及生物学特性测定。结果表明:引起红毛丹灵芝根腐病的病原菌为热带灵芝[Ganoderma tropicum(Jungh.)Bres.]。该病原菌菌丝生长最适温度为30℃,最适pH为6,黑暗条件有利菌丝生长,且在PDA培养基上生长较好。 |

In 2021, a root rot caused by Ganoderma was found on a living Nephelium lappaceum in Baoting botanical garden, Hainan Province. In order to identified the pathogen and clarify its biological characteristics, the author isolated 10 strains with the same morphology from its basidiocarp by conventional tissue isolation method, and selected the representative dominant strain (No. HMDGF001) for pathogenicity determination, morphological characteristics identification, rDNA-ITS, SSU and LSU sequence analysis, constructed phylogenetic tree and determined the biological characteristics. The results showed that the pathogen causing root rot of Nephelium lappaceum was Ganoderma tropicum (jungh.) Bres. The optimum temperature for mycelial growth of the pathogen is 30 ℃, the optimum pH is 6, the dark condition is conducive to mycelial growth, and it grows well on PDA medium. |
