以鳄梨组培苗为材料,在不同LED光质组合下,探讨对鳄梨组培苗生长和内源激素变化的影响。白光为对照,经过不同LED光质组合(1R1B、1B1G、1R1B1G)培养,诱导35天,分化100天,测苗高,增殖系数,采用LC-MS/MS进行测定,分析不同LED光质组合对鳄梨组培苗生长和内源激素含量变化的影响。结果表明,与对照相比,1R1B组增殖系数达5.64,苗高2.85cm,对四种生长素、六种细胞分裂素、GA3含量影响较大且促进;1B1G组增殖系数为2.87,苗高1.89cm,对五种生长素、九种细胞分裂素、ACC含量影响较大且促进;1R1B1G组增殖系数4.95,苗高2.33cm,对四种生长素、四种细胞分裂素影响较大且促进。LED光质可替代白光应用于鳄梨组培苗快速繁育,1R1B光质组合在相同培养时间内影响了多种内源激素的增加,协同作用刺激了组培苗的生长,更有利于快速繁殖。 |

To investigate the effects of different LED light quality combinations on the growth and endogenous hormone content of Avocado tissue culture seedlings,to provide a theoretical basis for the healthy Avocado tissue culture seedling rapidly. There were 4 treatment groups, Ⅰ-white(CK), Ⅱ-1R1B, Ⅲ-1B1G and Ⅳ-1R1B1G. Cultured for 135 days, to measure the seedling height and proliferation coefficient, and to analysis the endogenous hormone content by LC-MS/MS. Compared with white light, the proliferation coefficient of 1R1B group was 5.64 and the seedling height was 2.85cm, which had a great influence on the contents of four auxin, six cytokinins and GA3; The increment coefficient of 1B1G group was 2.87 and the seedling height was 1.89cm, which had a great influence on the contents of five auxin, nine cytokinin and ACC; The increment coefficient of 1R1B1G group was 4.95, the seedling height was 2.33cm, and it was sensitive to four auxin and four cytokinin. LED light quality could replace white light and apply to rapid propagation of avocado tissue culture seedlings. 1R1B light quality combination affected the increase of a variety of endogenous hormones in the same culture time. The synergistic effect stimulated the growth of tissue culture seedlings and was more conducive to rapid propagation. |
