      泛素结合酶变体蛋白(Ubiquitin-conjugating E2 enzyme variant, UEV),又被称为类泛素结合酶蛋白家族。 UEV参与激素信号、DNA 损伤和非生物胁迫等多种生物学功能,但尚未见其调控香蕉(Musa acuminata)果实成熟的研究报道。本研究在香蕉中共鉴定到13个UEV基因,MaUEV基因编码区域介于471 bp(MaUEV6)—1637 bp(MaUEV8),编码氨基酸数量介于127 aa(MaUEV6)—495 aa(MaUEV8),分子量介于14212.34 Da—56209.83 Da,13个MaUEV基因的等电点均小于7。8个MaUEV基因为稳定蛋白,5个MaUEV基因为不稳定蛋白,13个MaUEV基因均为亲水性蛋白。13个MaUEV基因均由α-螺旋、β-转角、扩展链结构、无规则卷曲组成。亚细胞定位分析发现除MaUEV3定位在叶绿体、细胞质和细胞核外,其他基因都定位在细胞核。对MaUEV基因结构分析发现,除了MaUEV4没有内含子外,其余12个MaUEV基因均由内含子和外显子组成。对MaUEV基因家族在香蕉后熟过程中的表达分析发现,MaUEV9表达量在香蕉后熟过程中逐渐下降并在成熟时显著下调,MaUEV13在香蕉后熟过程中逐渐增加并在成熟时表达量显著上调,据此推测MaUEV9 和MaUEV13在香蕉后熟过程中起着不同的作用。本研究可为后续深入阐明UEV基因家族不同成员在香蕉后熟过程中的作用奠定基础。
英文摘要:
      Ubiquitin-conjugating E2 enzyme variant (UEV), also known as ubiquitin like binding enzyme protein family. UEV participates many biological functions such as hormone signaling, DNA damage and abiotic stress. However, there was no research report on UEV gene and banana (Musa acuminata) fruit ripening. In this study, 13 UEV genes were identified in banana. The coding region of MaUEV gene were from 471bp (MaUEV6) to 1637bp (MaUEV8). The number of amino acids were from 127 aa (MaUEV6) to 495 aa (MaUEV8). The molecular weights were from 14212.34 Da to 56209.83 Da. The isoelectric points of all MaUEV genes were less than 7. Eight MaUEV were stable proteins and five MaUEV were unstable proteins. All MaUEV genes were hydrophilic proteins, and were composed of alpha helix, beta turn, extended strand, random coil. Subcellular localization analysis showed that all genes were located in nucleus, except MaUEV3 was located in chloroplast, cytoplasm and nucleus. The structural analysis of MaUEV genes showed that 12 MaUEV genes were composed of intron and exon, but MaUEV4 had no intron. The expression of MaUEV gene family was analyzed during banana ripening. MaUEV9 expression was gradually decreased during banana ripening, and significantly decreased at maturity. While MaUEV13 expression was gradually increased during banana ripening, and significantly increased at maturity. Therefore, it was speculated that MaUEV9 and MaUEV13 played different roles in banana ripening. This study will provide a foundation for further clarifying the role of different members of UEV gene families in bananaripening.
