[目的]为明确重庆市长寿区柑桔砂皮病菌的生理生化特征及生长条件,筛选新型高效纳米药剂。从重庆市长寿区柑桔果园采集柑桔砂皮病样品,分离纯化菌株,应用形态学和rDNA-ITS序列分析鉴定病原菌,并研究其生物学特性;采用菌丝生长速率法测定3种纳米试剂及3种抑菌剂的抑菌率,测定其对柑桔砂皮病菌的毒力。[结果]鉴定获得柑桔砂皮病菌菌株1株,不同培养基培养的菌株生长性状有明显差异,其对有机碳源甘油、麦芽糖、可溶性淀粉的利用效果较佳,菌丝扩展速度较快,对有机氮源蛋白胨的利用效果最好。菌株在15~28 ℃范围内均能生长,最适生长温度20~28 ℃,致死温度35 ℃,pH值5~9均能生长,最适pH值5~7;菌株只在光暗交替培养条件下才出现花瓣状轮纹。吡唑醚菌酯? 喹啉铜、噻呋?氟环唑、戊唑醇对柑桔砂皮病菌具强抑制作用,EC50范围在0.01174~0.1128 μg/mL。SiO2纳米试剂、土霉素碳量子点(OTC-CQDs)纳米试剂和星状阳离子聚合物(SPc)纳米载体皆对砂皮病菌具有较强的抑制作用。其中,OTC-CQDs纳米试剂的抑制作用最强,EC50为0.02243 μg/mL,SPc的抑制作用最弱,EC50为0.7907 μg/mL。 |

[Objective]In order to clarify the physiological and biochemical characteristics and growth conditions of citrus melanose in Changshou District of Chongqing, new high-efficient nanoagents were screened. Samples of citrus melanose were collected from citrus orchards in Changshou District of Chongqing, and the strains were isolated and purified.The pathogens were identified by morphology and rDNA-ITS sequence analysis, and their biological characteristics were studied. The bacteriostatic rate of three nanoreagents and three fungicides were determined by mycelium growth rate method, the virulence of citrus melanose was determined. [Results] One strain of citrus melanose was identified, and the growth characteristics of the strains cultured in different media were significantly different. The strains had the best utilization effect on organic carbon sources such as glycerol, maltose and soluble starch, the fastest mycelial growth rate, and the best utilization effect on organic nitrogen source peptone. The strain could grow at 15 ~ 28 °C, the optimum growth temperature was 20 ~ 28 °C, the lethal temperature was 35 °C, the pH value was 5 ~ 9, the optimum pH value was 5 ~ 7. The strain only showed petal-like whorls under alternating light and dark culture conditions. Pyraclostrobin quinolate?copper, Thifluzamide? epoxiconazole,Tebuconazole had strong inhibitory effect on citrus melanose , and the EC50 value ranges from 0.01174 to 0.1128 μg/mL.SiO2 nanoreagents, totetracycline carbon quantum dots (OTC-CQDs) nanoreagents and astrocationic polymer (SPc) nanocarriers all have strong inhibitory effects on citrus melanose. Among them, the inhibition effect of OTC-CQDs nanoreagents was the strongest, EC50 value was 0.02243 μg/mL, SPc was the weakest, and EC50 value was 0.7907 μg/mL. |
