为建立南高丛蓝莓明星离体再生体系,以带腋芽嫩枝条为外植体试材,诱导腋芽萌发获得无菌材料的叶片为离体培养试验材料,研究了灭菌时间、不同浓度和种类的植物生长调节剂对外植体灭菌、不定芽的诱导和继代增殖培养的影响。结果表明:外植体最佳灭菌方法为75%酒精浸泡1Os,再用0.1%升汞溶液灭菌5-6min;腋芽萌发最适培养基为PM+2.0mg?L-1ZT;不定芽诱导最适培养基为WPM+0.5 mg?L-1TDZ+0.2 mg?L-1IBA;不定芽增殖培养最适培养基为WPM+2.5 mg?L-1ZT。 |

In order to establish the invitro regeneration system of Southern highbush blueberry Star,the tender branches with axillary buds were used as explants, Aseptic materials obtained by inducing axillary bud germination.The research contents include effects of sterilization time, different concentrations and kinds of plant growth regulators on the sterilization of explants, induction of adventitious buds and subculture proliferation.The results showed that:The best sterilization method for explants is to soak them in 75% alcohol for 1Os, 0.1% mercuric chloride for 5-6 min.The optimum medium for axillary bud germination is WPM+2.0mg ? L-1ZT, for adventitious bud induction is WPM+0.5 mg ? L-1TDZ+0.2 mg ? L-1IBA,and adventitious bud proliferation is WPM+2.5 mg ? L-1ZT. |
