      柑橘衰退病毒(Citrus tristeza virus,CTV)是对柑橘产业具有毁灭性危害的病原之一。为了明确JY基因型分离株在我国的发生及其分子特征,本研究对4个地区的野生柑橘和9个柑橘产区的栽培柑橘进行JY基因检测,并克隆测序JY 基因型分离物的ORF1b、p33、p25、p23基因进行分析。结果显示仅来自湖南江永自然生态中的46个道县野橘样品中检出12个阳性样品,说明JY 基因型在我国的发生分布并不广泛。ORF1b、p33、p25、p23基因序列多样性、遗传分化、分子变异分析表明,JY 基因型12个分离物间核苷酸和氨基酸序列相似性极高(>96.8%),但JY 基因型分离物与CTV其他基因型分离物存在明显的遗传分化:ORF1b、p33、p25、p23基因序列的平均核苷酸和氨基酸序列相似性分别为79.2%-92.7%、80.8%-85.9%、90.5%-93.6%、87.5%-91.8%和89.5%-98.1%、80.8%-84.4%、93.7%-97.3%、85.4%-91.6%;Fst均明显高于0.25;CTV JY基因型种群与其他基因型种群的差异非常大(分子变异大于43%,P值为0.001),且基因交流小(Nm=0.396)。ORF1b、p33、p25、p23基因单独及4个基因联合系统发育分析,表明JY 基因型12个分离物均与JY-2 聚集在同一簇,且独立于现有CTV基因型之外,进一步证实JY 基因型与CTV其他基因型分离物存在明显的遗传分化,可能为我国特有CTV基因型株系。
英文摘要:
      Citrus Tristeza virus (CTV) is one of the devastating phytopathogens that threaten the citrus industry. This study conducted to clarify the presence of JY genotype isolates and their molecular characteristics in China by detecting JY genes in wild citrus from four regions and cultivated citrus from nine citrus-producing areas, as well as cloning and sequencing ORF1b, p33, p25, and p23 genes from isolated JY genotypes. The results demonstrated that only 12 positive samples were detected among 46 Dao County wild orange samples from the natural ecology of Jiangyong, Hunan. This indicates that the JY genotype is rarely found in China. Sequence diversity, genetic differentiation, and molecular variation analyses of ORF1b, p33, p25, and p23 genes revealed a high degree of similarity between the 12 isolates of the JY genotype (> 96.8%). Nevertheless, there was a significant genetic divergence between the JY genotype isolates and the other CTV genotype isolates. The average nucleotide and amino acid sequence similarities of ORF1b, p33, p25, and p23 gene sequences were 79.2%-92.7%, 80.8%-85.9%, were all significantly higher than 0.25. CTV JY genotype populations had high molecular variability (> 43%, p-value 0.001) and low gene exchange (Nm=0.396). A phylogenetic analysis of the ORF1b, p33, p25, and p23 genes alone and in combination revealed that all 12 isolates of the JY genotype clustered with JY-2 and were independent of the existing CTV genotypes. This confirms that the JY genotype is genetically distinct from other CTV genotype isolates in China and maybe a unique strain of the CTV genotype.
