      为探明欧亚葡萄“魏可”在果皮转色过程中转录组水平的变化,于果皮转色期、转色后15天以及完熟期采集果实样品,利用RNA-Seq测序技术,对转色过程中相关基因表达量的变化规律进行探讨。结果得到葡萄转色不同阶段在转录组水平上存在显著差异表达基因。转色后15天vs转色期、完熟期vs转色期以及完熟期vs转色后15天之间的显著差异基因数分别为3096个、4444个和 1156个,且均含有显著上调和显著下调基因。 GO富集分析表明,转色后15天vs转色期在整体上以及下调差异表达基因间、完熟期vs转色期在整体上以及上下调差异表达基因间、完熟期vs转色后15天在上调差异表达基因间存在显著富集过程。鉴定出KEGG富集的代谢通路有10条,分别在转色后15天vs转色期下调差异表达基因、完熟期vs转色期上下调差异表达基因、完熟期vs转色后15天下调差异表达基因之间,主要涉及光合作用和半乳糖代谢途径。转录因子分析共鉴定出62个转录因子基因家族的446个转录因子。欧亚葡萄“魏可”果皮转色过程中转录组水平上相关基因表达量存在显著差异,且GO和KEGG注释分析均鉴定出显著富集的过程及代谢途径。为进一步大量挖掘与“魏可”葡萄果皮转色相关重要表达基因奠定一定的数据基础和科学参考。

      It is important to explore the changes during veraison of Vitis vinifera ‘Wink’ at transcriptome level. The fruit samples were collected at before color-changed, 15 days after color-changed and full ripe stage, respectively. And RNA-sequencing technology was used to explore the changes of relative gene expression during veraison. Significant different expression genes (DEGs) were detected by RNA-sequencing technology. The analysis revealed 3096 significant DEGs of 15 days after color-changed vs before color-changed, 4444 significant DEGs of full ripe stage vs before color-changed, and 1156 significant DEGs of full ripe stage vs 15 days after color-changed. Up-regulated and down-regulated DEGs were both detected among the three comparing. Through Go enrichment analyzing, some significant enrichment processes were detected, which involved total analysis and down-regulated DEGs of 15 days after color-changed vs before color-changed, total analysis and up/down-regulated DEGs of full ripe stage vs before color-changed, up-regulated DEGs of full ripe stage vs 15 days after color-changed. Ten metabolic pathways significantly enriched in KEGG pathways were identified and distributed among down-regulated DEGs of 15 days after color-changed vs before color-changed, up/down-regulated DEGs of full ripe period vs before color-changed, down-regulated DEGs of full ripe stage vs 15 days after color-changed. These significant pathways were mainly involved photosynthesis and galactose metabolism. Transcription factor analysis identified 446 transcription factors of 62 transcription factor gene families. There were some significant differences in the expression of related genes during the process of ‘Wink’ veraison. Go and KEGG annotation analysis have both identified the significant enrichment process and metabolic pathway. It lays a certain data foundation and a scientific reference for further mining the important expression genes related to ‘Wink’ grape veraison.
