本研究利用RNAi介导的病毒抗性的原理,以柑橘上发生频繁、危害严重的柑橘黄化脉明病毒(Citrus yellow vein clearing virus,CYVCV)为研究对象,以期得到对CYVCV具有抗性的转基因植株。根据CYVCV的CP基因和RP基因序列设计含有不同酶切位点的特异性扩增引物,获得RNA干扰片段,分别构建RNA干扰载体ihpCP-pGN、ihpRP-pGN和ihpCPRP-pGN,将干扰载体转入农杆菌EHA105后,采用农杆菌介导法进行柑橘的遗传转化,获得两株转基因阳性植株,为培育抗CYVCV病毒柑橘品种提供技术参考和理论基础。 |

In this study, based on the principle of RNA interference mediated viral resistance, Citrus yellow vein clearing virus (CYVCV), which occurs frequently and is highly harmful in citrus, was taken as the research object in order to obtain CYVCV resistant transgenic plants. According to the CP gene and RP gene sequences of CYVCV, specific amplification primers containing different enzyme restriction sites were designed to obtain RNA interference fragments. RNA interference vectors ihpCP-pGN, ihpRP-pGN and ihpCPRP-pGN were constructed respectively. After the interference vectors were transferred into Agrobacterium EHA105, Two transgenic positive plants were obtained by Agrobacterium-mediated genetic transformation of citrus, which provided technical reference and theoretical basis for cultivating CYVCV virus resistant citrus varieties. |
