以‘粉玉1号’草莓匍匐茎芽为试料,通过外植体消毒、茎尖剥离、不定芽诱导、生根培养,建立了‘粉玉1号’草莓茎尖脱毒及快繁体系。结果表明,‘粉玉1号’草莓茎尖最佳的灭菌方法为75%乙醇处理30s,再用0.1%升汞溶液处理10 min;初代培养基以MS + 0.5 mg·L-1 6-BA为最佳;生根培养基为不加激素的1/2 MS为宜。同时利用RT-PCR技术对‘粉玉1号’茎尖培养苗进行了草莓皱缩病毒、草莓斑驳病毒、草莓轻型黄边病毒和草莓镶嵌病毒4种病毒的检测,未发现病毒感染。本研究建立的茎尖脱毒及快繁体系可为‘粉玉1号’草莓的进一步开发利用以及工厂化育苗提供有益参考。 |

With the stem tip of ‘Fenyu No.1’strawberry as the test material, the virus-free and rapid propagation system of ‘Fenyu No.1’ strawberry was established through explant disinfection, stem tip stripping, adventitious bud induction and rooting culture. The results showed that the best sterilization treatment of ‘Fenyu No.1’ strawberry stem tip was 75% alcohol for 30s, 0.1% mercury chloride for 10min. MS with 0.5 mg·L-1 6-BA was the optimum medium for primary culture. The rooting medium should be 1/2 MS without any hormone. Meanwhile, RT-PCR was employed to detect 4 common viruses present in the plantlets of ‘Fenyu No.1’, and no virus was detected. The stem tip virus-free and rapid propagation system established in this study will provide a useful reference for the further development and utilization of ‘Fenyu No.1’ strawberry and the industrialized seedling breeding. |
