以不同倍性草莓资源的嫩叶为材料,根据待测样品倍性预估基因组大小后分别选择水稻日本晴、番茄、玉米B73作为内参,利用流式细胞仪测定样品的基因组大小。不同草莓资源的基因组大小分别为:二倍体森林草莓(233±0.78)Mb、绿色草莓(234±0.82)Mb;四倍体西南草莓(492±2.12)Mb、伞房草莓(雄株)(488±7.07)Mb、伞房草莓(雌株)(491±4.95)Mb;六倍体麝香草莓(644±1.73)Mb;八倍体栽培草莓品种‘章姬’(793±7.78)Mb、‘蒙特瑞’(761±0.00)Mb;十倍体野生资源择捉草莓(938±6.36)Mb、十倍体栽培品种‘桃熏’(1007±6.36)Mb。 |

The young leaves of different ploidy strawberry resources were used as materials, and the genome sizes of the samples were determined by flow cytometry after predicting the genome sizes according to the ploidy of the samples to be tested and selecting rice , tomato and maize as internal reference, respectively. The genome sizes of different Fragaria resources were as follows: Diploid, F. vesca (233±0.78) Mb, F. viridis (234±0.82) Mb; Tetraploid, F.moupinensis (492±2.12) Mb, F. corymbosa (male) (488±7.07) Mb, F. corymbosa (female) (491±4.95) Mb; hexaploid, F. moschata (644±1.73) Mb; Octoploid cultivars, ''Zhangji'' (793±7.78) Mb, ''Monterey'' (761±0.00) Mb; Decaploid wild resource, F. iturupensis (938±6.36) Mb, Decaploid cultivar ''Tokun'' (1007±6.36) Mb. |
