为探究水分胁迫对‘赤霞珠’葡萄果实品质及有机酸合成相关基因表达的影响。以9年生‘赤霞珠’葡萄为试材,在花后18 d,开始进行无胁迫(对照)、设置T1(水涝胁迫)和T2(干旱胁迫),测定了葡萄可滴定酸含量、果百粒重、可溶性固形物、单宁、总酚和有机酸含量。采用实时荧光定量PCR技术检测有机酸合成相关基因的表达量。与对照相比,在花后85~105 d时,T1可提高IDH、PEPC和MDH的基因表达量,T1处理下PEPC基因表达量在各生长阶段均有所上升,在花后25~45 d时,T2处理抑制了IDH和CS基因表达量。同时,葡萄从坐果期至成熟期,在T1处理下可提高百粒重、可滴定酸和有机酸含量,降低可溶性固形物、单宁和总酚含量。适度的水分胁迫可以提高果实品质和有机酸含量,还为贺兰山东麓赤霞珠葡萄栽培管理和水分高效利用提供理论依据。 |

To explore the effect of water stress on fruit quality and gene expression of organic acid synthesis of Cabernet Sauvignon grape. Using the 9-year-old "Cabernet Sauvignon" grape as the test material, no stress was 18 d (control), T1 (waterlogging stress) and T2 (drought stress). Expression level of genes involved in organic acid synthesis was measured by quantitative real-time PCR. Compared with control, T1 increased the gene expression of IDH, PEPC and MDH at 85 and 105 d after flower, and PEPC gene expression increased at all growth stages under T1 treatment, and T2 treatment at 25 to 45 d after flower. At the same time, the content of grain weight, titrated acid and organic acid, and the content of soluble solid substance, tannin and total phenols can be reduced under T1 treatment. Moderate water stress can improve fruit quality and organic acid content, and also provide a theoretical basis for the grape cultivation management and efficient water utilization at the eastern foot of Helan Mountain. |
