为筛选火龙果组织培养适宜的外植体类型、消毒方法及适宜的不定芽的诱导方法,为火龙果的快速繁殖提供技术支持。对外植体类型、灭菌时间、消毒药剂及激素配比对不定芽的诱导等进行了研究,结果表明:(1)3种类型外植体中最容易消毒的是种子。(2)升汞对 3 种外植体的消毒效果明显优于次氯酸钠。(3)3种外植体最适宜的消毒方法分别为:种子0.1%升汞消毒3min,近成熟茎0.1%升汞5 min+0.1%升汞7min,幼嫩茎段0.1%升汞5min+ 0.1%升汞3min,其存活率分别为94.44%,83.3%,75%。(4)近成熟茎段诱导不定芽效果最好的培养基组合为MS+6-BA5.0 mg/L + NAA 0.1 mg/L。诱导率为77.78%。幼嫩茎段的最佳不定芽诱导培养基为 MS+3.0 mg/L 6-BA+0.2 mg/L NAA,诱导率为83.33%。最适合种子幼茎增殖的培养基配比为MS+3.0 mg/L 6-BA+0.2 mg/L IAA。 |

In order to screen the suitable explant type, disinfection method and suitable adventitious bud induction method for tissue culture of pitaya, and provide technical support for the rapid propagation of pitaya. The effects of explant types, sterilization time, disinfectants and different plant growth regulators on the induction of adventitious buds were studied.The results showed that: (1) the seed was the most easily disinfected among the three types of explants. (2)The disinfection effect of mercuric chloride on the three explants was significantly better than that of sodium hypochlorite.(3)The most suitable disinfection methods of the three explants were : seed 0.1 % mercuric chloride for 3 min, near mature stem 0.1 % mercuric chloride |
