      为进一步提高平邑甜茶的离体快繁效率,本研究以平邑甜茶新梢带芽茎段为外植体,探究不同外植体消毒方式、不同的激素配比对组培苗增殖和生根培养的影响,建立平邑甜茶离体快繁体系。结果表明:平邑甜茶的新梢茎段用75%酒精处理30 s+2%次氯酸钠溶液处理12 min的消毒效果较好,成活率达81.11%。最佳增殖培养基为MS+1.0 mg/L 6-BA+0.1 mg/L IBA,增殖系数为7.09;最适生根培养基为1/2MS+0.15 mg/L NAA,该处理下的生根率为75.55%,平均根系数量和长度分别为3.23条、3.57 cm;生根后的组培苗在蛭石:珍珠岩:营养土质量比为1:1:1基质中的移栽成活率为90%。本研究成功建立平邑甜茶离体快繁体系,为平邑甜茶砧木的育苗提供可靠的理论依据。
英文摘要:
      In order to further improve the efficiency on tissue culture and rapid propagation system of ‘Malus hupehensis’, the stem segments with buds of ‘Malus hupehensis’ were used as explants to explore the effects of different explants disinfection methods as well as different hormone ratios on the proliferation and rooting culture of tissue culture seedlings. The results showed that the explants been immersed in 75% alcohol for 30 seconds, and then in 2% sodium hypochlorite for 12 mins was the preferable method of disinfection, the survival rate was 81.11%. The optimal proliferation medium was MS+1.0 mg/L 6-BA+0.1 mg/L IBA, with a proliferation coefficient of 7.09. In addition, the optimal rooting medium was 1/2 MS+0.15 mg/L NAA and the rooting rate was 75.55%, and the average number and length of roots were 3.23 and 3.57 cm respectively. After 30 days of rooting, the survival rate was 90% of the rooted bud was transplants to vermiculite: perlite: nutrient soil = 1:1:1. Herine, the apomictic apple rootstock ‘Malus hupehensis’ tissue culture and rapid propagation system was established, that would provide reliable theoretical basis for the seedling raising of ‘Malus hupehensis’.
