      采用高通量测序技术对化州柚Citrus grandis ‘Tomentosa’叶绿体基因组进行测序,使用生物信息学方法对其进行组装、注释以及解析。结果显示,化州柚叶绿体基因组为160 139 bp的环状双链四分体,由87 733 bp的大单拷贝区、18 410 bp的小单拷贝区以及穿插两者之间的两个26 998 bp的反向重复区构成,并且含有133个基因。化州柚叶绿体基因组共有26 244个密码子,其中以A/T碱基结尾的密码子居多,且检测到的104个简单重复序列中,以A/T碱基的单核苷酸重复基序为主。化州柚等植物叶绿体基因组的单拷贝区/反向重复区边界构成和全序列相似度较高,变异碱基主要分布在trnS-trnG、petN-psbM、psbC-trnS、trnT-trnL、accd-psaI、petA-psbJ、rps3-rpl22、ndhF-rpl32、ccsA-psaC和ycf1等基因间隔区或基因。系统进化分析显示化州柚与同属植物柚、甜橙的叶绿体基因组同源性最高。
英文摘要:
      In this study, the whole chloroplast genome of Citrus grandis ‘Tomentosa’ was sequenced using high-throughput sequencing technology and successively analyzed with bioinformatics approaches. The results demonstrated that the plastome of Citrus grandis ‘Tomentosa’ was a circular molecule with a size of 160 139 bp. The entire chloroplast genome embodied a 87 733 bp of large single copy region and a 18 410 bp of small single copy region, which were compartmentalized by two 26 998 bp of inverted repeat. Among 133 genes annotated from the plastome, 88 were characterized as protein-coding genes, 8 belonged to rRNA genes and the rest of them were tRNA genes. Codon preference and simple repeat sequence were also studied to uncover the characterization of the whole plastome of Citrus grandis ‘Tomentosa’. The comparative analysis among 10 Citrus species revealed ten divergence regions including trnS-trnG, petN-psbM, psbC-trnS, trnT-trnL, accd-psaI, petA-psbJ, rps3-rpl22, ndhF-rpl32, ccsA-psaC and ycf1 were identified as potential DNA barcoding for species identification in Citrus genus. Phylogenetic analysis based on the full-length chloroplast genomes of 20 species uncovered Citrus grandis ‘Tomentosa’ is a sister taxon to C. sinensis and C. maxima.
