      品种鉴定是植物品种保护的前提。基于表型性状的DUS测试是受国际认可和具有法律依据的鉴定方法;基于分子标记的检测则能准确提示品种的遗传特征。对果树作的完整DUS测试,测试者往往面临长测试周期和近似品种选择困难的挑战。本研究首先对5个测试材料进行部分DUS测试,结果表明它们之间无明显性状差异。测试材料与无核金诺在果实重量性状上有一个代码的差异,与091无核沃柑在4个性状上分别有1-2个代码的差异,因此确认091无核沃柑(Citrus reticulata Blanco)和无核金诺(C. reticulata Blanco)为测试材料的近似品种。进一步利用33个适用于琼脂糖电泳检测InDel标记进行分子检测,结果表明5个测试材料之间无位点差异,测试材料与无核金诺和091无核沃柑分别有0和12个位点的差异。综合上述两种方法所得结果,这5个测试材料被鉴定为无核金诺。本研究为准确快速鉴定柑橘品种提供解决有效方案。
英文摘要:
      Precise variety identification is a prerequisite for plant variety protection. DUS (distinctness, uniformity, and stability) testing based on phenotypic traits is an internationally recognized and legally valid method,and the testing based on molecular marker can accurately reveal the genetic characteristics. For the complete DUS testing of fruit crops, testers are usually faced with challenges due to the long testing period and dilemma in selecting similar varieties. In this study, we first performed partial DUS testing for five mandarin samples, and they had no visible differences in traits. These samples had one-code discrepancy of fruit weight with “KinnowLS” mandarin but one to two-code discrepancies of four traits with “091 Seedless Orah” mandarin, indicating that “091 Seedless Orah” mandarin (Citrus reticulata Blanco) and “KinnowLS” mandarin (C. reticulata Blanco) were the similar varieties. Furthermore, 33 agarose gel electrophoresis-resolved InDels markers were applied for the molecular testing. There were no polymorphic discrepancies among the five samples and “KinnowLS” mandarin, but the tested samples showed 12 different loci compared to “091 Seedless Orah” mandarin. Taken together, the five samples were identified as “KinnowLS” mandarin. This study offers an effective solution for the quick and precise variety identification of citrus crops.
